circRNA过表达

circRNA由于结构的特殊性,与miroRNAlncRNA有着很大差异,让研究者对于它的功能的研究也是难于开展。吉赛生物经过不断设计、验证和优化,先后测试了30多种circRNA表达框架,优化出了特异性准确过表达circRNA分子的载体。载体含有表达circRNA的表达框,可以在细胞内通过RNA剪接形成circRNA分子;广大研究者可以将目标circRNA的线性序列PCR扩增出来,直接连接到载体上生成目标circRNA分子。

 

过表达载体

过表达效率高:circRNA表达载体上加有吉赛生物专利技术的上下游环化框架,验证过的所有 circRNA都能显著过表达50倍以上,最高可过表达800倍。

序列环化准确:环化框架内部引入吉赛生物专利技术的环化介导序列,有效保证目标 circRNA的线性序列准确环化,验证过的所有circRNA都能准确环化,无碱基添加或缺失。

过表达稳定性高:验证过的目标 circRNA200nt2500nt序列都能实现准确高效过表达。

适用范围广:pCD5-ciR、pCD25-ciR 可转染细胞瞬时表达,pLO5-ciR、pLC5-ciR可转染细胞瞬时表达,也可包装慢病毒构建稳定细胞系,pLC5-ciR可同时表达 GFP 荧光和 Puromycin 抗性基因,可以方便地进行筛选。
